no, the stone of destiny can not be used on already awakened classes, it is used only at the moment of awakening as part of the awaken quest...i.e you have level 85 cardinal - start awaken quest - at point where you get to chose your path, instead of having only one option (healer) you are able to select from all 4th class options.
